Everything depends on how much time you are prepared to spend on the Route.

Most people do the full trip, Cape Town to Durban, or the other way around.
Personally, I would advise that you start in CT and get your flight into CT and you get to see Table Mountain as you come into land - really an amazing sight.

If you plan on about 4-5days in Cape Town, you can climb Table mountain, do a shark cage dive, a wine tour or two in the surrounding area, then maybe another day spent at one of the amazing beaches (eg. Camps Bay.)
If you are staying in CT most of the backpackers there are reasonably cheap if you are willing to share a dorm with 3-4 others and even private rooms are cheap too ranging from R100-200 (R100 is the normal price for dorm rooms.) I reccomend any backpackers in Long Street, it is right in the center of the city, and has a great night life and a lot of backpackers to choose from, my personal favourite is Long Street Backpackers - the cheapest rooms but the best location on the street, as well as the best atmosphere.
I advise starting in Cape Town first purely because its a classic start point and you can meet up with new people there and start heading up the garden route with your new friends. Plans are always changing on the route. You can take busses, or hire cars, or even short internal flights. The Baz Bus is very popular, you can buy a few month ticket and use it as much or little as you like, and they drop you door to door from one backpackers to the next.

In the end I reckon the best you can do is to go there, have a general time frame you want to do everything in, and ask along the way, you will meet people coming the other way, they will tell you the best places they seen or places to avoid. With travelling its the people you meet that makes the difference, and thats what makes the garden route so amazing, you meet new people and do amazing things.

Once you arrive, most backpackers have a free book which you can just ask for or will be on display called "Coast to Coast" this will give you all the information you will need. Asking the managers at places you stay is also a great idea, they often know the other managers along the route so they can tell you the best places at the time, and also may be able to pre book you into other places.

Whatever you do, just enjoy and have fun!
